Prostaglandin E2 (PGE2) is known to suppress collagen expression in fibroblasts through activation of cAMP-dependent protein kinase A (PKA). However, the actual mechanism by which PGE2-PKA signaling inhibits collagen expression in fibroblasts has never been clarified. In this study, we found that PGE2 inhibited protein translation through PKA in normal human lung fibroblasts, thus suppressing not only collagen production but also global protein synthesis. Similar effects of PGE2 were observed in mouse peritoneal macrophages. These findings identify inhibition of translation as a new mechanism for PGE2 regulation of cellular function.
