Tumor heterogeneity from genetic instability in cancer cells is one of the major reasons for treatment failure. Cancer cells adapted to environmental and therapeutic insults survive and expand in the human body, which is analogous to the “evolution” in the ecological system on the planet. Excessive replication of DNA plays a critical role in genetic instabilities including gene amplification and genomic rearrangements. We found that low-fidelity Y-family polymerases are involved in this aberrant replication. This finding may open a new avenue to cancer therapeutics.
